The respiratory physiology in 35 pro mille artificial seawater has been studied for four strains of adult <i>Artemia</i> . The scope of activity and the potential activity range were studied by measuring the respiratory rate of actively swimming and anaesthetized animals and the respiration of total homogenates. The experiments were carried out at temperatures ranging from 5 to 45°C. The lower lethal value for male and female <i>Artemia</i> from the four strains (Great Salt Lake, Macau and Sfax) being 5°C. A flow-through test system for the study of toxicity and bioaccumulation processes is described. The apparatus can operate within a very wide range of flow rates. Flow deviations are less than 2% and the variation in test-agent concentration, (determined for copper) ranges from 3 to 8%. As a practical test a copper-toxicity experiment on adult <i>Artemia</i> was carried out and the results compared with data obtained from a similar but static assay. The effect of salinity on cadmium uptake by the brine shrimp <i>Artemia fransiscana</i> was studied in chemically defined saltwater solutions. Shrimp were acclimated to different salinities and exposed to the metal in solutions of differing salinity and composition. Within each acclimation group, the cadmium uptake decreased with increasing salinity of exposure. Most of this variation was explained by changes in the activity of the free metal ion. Among the acclimation groups, uptake increased by increasing salinity of acclimation. Most of this variation was explained by changes in the permeability of the shrimp. The effect of temperature on the uptake of copper by the brine shrimp <i>Artemia franciscana</i> has been studied in chemically defined saltwater solutions. Animals were acclimated to temperatures ranging from 10°C to 35°C and exposed to copper at different temperatures within this range. The results show that within each temperature acclimation group, copper uptake increases with increasing temperature of exposure. Within each temperature acclimation group most of the variation can be related to the combined effect of temperature on the chemical speciation and diffusion rate of copper in the diffusion layer lining the exchange surfaces. Bioavailability of oxygen for the saline-water invertebrate <i>Artemia franciscana</i> was studied, since both oxygen concentration and oxygen diffusion rate change with salinity. Total haemoglobin concentration and the relative contribution of each of three haemoglobins was measured in specimens acclimated to different salinities and oxygen concentrations. Both haemoglobin concentration and contribution were influenced by salinity and the group observed (males, females with and females without eggs). A combined gamma scintillation/liquid scintillation technique for the sequential determination of two radioactive tracers is described. The method was developed using a combined gamma/ß-emitter (<sup>57</sup>Co) and a pure ß-emitter (<sup>45</sup>Ca). First, the <sup>57</sup>Co radioactivity was determined by counting the samples in a gamma scintillation analyzer in the energy region 80-165 keV. Next, the samples were counted in a liquid scintillation analyzer. Only one energy region was used to count both isotopes to maximize the counting efficiencies. The temperature in a natural <i>Artemia</i> habitat usually shows diurnal cyclic variations. It is important for inoculation and culturing purposes to know the survival under such conditions. The survival of four <i>Artemia</i> strains, Great Salt Lake and San Francisco Bay (USA), and Sfax and Mégrine (Tunisia) was studied over a period of 7 days, in different conditions of constant or diurnal fluctuating temperatures at two salinities. The effect of organic complexation, salinity, and pH on the uptake and toxicity of copper in <i>Artemia</i> was studied. The results were compared with data on the chemical speciation and lipid solubility of copper in saline solutions.The uptake experiments took 90 min and the toxicity tests 24h. A computer model was used to calculate the saline solution equilibrium speciation. The lipid solubility of copper was determined in a series of saline/olive oil partition tests.Complexation of copper by EDTA reduced the uptake and toxicity of copper.
